mod kr\u00e6ftceller. Det er principielt det, som forskere har udviklet, og som beskrives i denne artikel.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">De har lavet en ny medicin, FL1-PNU, der virker som et missil, der specifikt rammer et protein kaldet uPAR, som findes p\u00e5 kr\u00e6ftceller i bugspytkirtlen. Medicinen er testet i laboratoriet og p\u00e5 mus med <a href=\"https:\/\/jegharkraeft.dk\/bugspytkirtelkraeft\"><strong>kr\u00e6ft i bugspytkirtlen<\/strong><\/a>.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Resultaterne viser, at medicinen er effektiv til at dr\u00e6be kr\u00e6ftceller og f\u00e5 kr\u00e6ftsvulsten til at skrumpe. Samtidig hj\u00e6lper den med at &#8216;booste&#8217; kroppens eget immunforsvar, s\u00e5 det bedre kan bek\u00e6mpe kr\u00e6ften.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Medicinen er stadig p\u00e5 et tidligt stadie, men resultaterne er lovende og giver h\u00e5b om en ny og effektiv behandling mod kr\u00e6ft i bugspytkirtlen i fremtiden.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><span class=\"ez-toc-section\" id=\"Baggrund\"><\/span>Baggrund<span class=\"ez-toc-section-end\"><\/span><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">PDAC (kr\u00e6ft i bugspytkirtlen) er en aggressiv kr\u00e6ftform med en d\u00e5rlig prognose, prim\u00e6rt p\u00e5 grund af dens t\u00e6tte, bindev\u00e6vsagtige st\u00f8ttev\u00e6v, der hindrer effektiv l\u00e6gemiddellevering og immuncelle infiltration.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">ADC&#8217;er (m\u00e5ls\u00f8gende medicin), der leverer m\u00e5lrettet cytotoksisk (celledr\u00e6bende) behandling via tumor-specifikke antistoffer, har vist lovende resultater i behandlingen af \u200b\u200bforskellige solide (faste) tumorer. uPAR (et protein) er et attraktivt m\u00e5l i PDAC p\u00e5 grund af dets h\u00f8je forekomst i b\u00e5de tumorceller og st\u00f8ttev\u00e6vsceller.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><span class=\"ez-toc-section\" id=\"Studiemetoder\"><\/span>Studiemetoder<span class=\"ez-toc-section-end\"><\/span><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>ADC design og generering<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">T\u00e6nk p\u00e5 det som at bygge en lille pakkebombe, der kun rammer kr\u00e6ftceller. Forskerne tog et antistof (FL1), der kan genkende og binde sig til kr\u00e6ftceller, og koblede det sammen med en kraftig cellegift (PNU-159682). Forbindelsen mellem antistoffet og cellegiften er en slags &#8216;snor&#8217;, der f\u00f8rst &#8216;spr\u00e6nger&#8217; og frigiver giften, n\u00e5r pakken er kommet ind i kr\u00e6ftcellen.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>In vitro evaluering<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">In vitro betyder &#8220;i glas&#8221;, alts\u00e5 i laboratoriet.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Forskerne testede medicinen p\u00e5 forskellige kr\u00e6ftceller fra mennesker og mus i petrisk\u00e5le for at se, hvor effektiv den var til at dr\u00e6be dem.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">De unders\u00f8gte ogs\u00e5, om medicinen kunne dr\u00e6be kr\u00e6ftceller, der ikke havde det protein (uPAR), som antistoffet binder sig til, ved en slags &#8216;dominoeffekt&#8217; (bystander drab).<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>In vivo evaluering<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">In vivo betyder &#8220;i levende organismer&#8221;, alts\u00e5 i mus.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Forskerne testede medicinen p\u00e5 mus med kr\u00e6ft i bugspytkirtlen for at se, om den kunne bremse kr\u00e6ftens v\u00e6kst.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">De unders\u00f8gte ogs\u00e5, hvordan medicinen p\u00e5virkede musenes immunforsvar, og om den kunne hj\u00e6lpe immunforsvaret med at bek\u00e6mpe kr\u00e6ften.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Flowcytometri og immunhistokemi<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Det er to metoder, som forskerne brugte til at unders\u00f8ge cellerne i musene n\u00e6rmere. Flowcytometri er en teknik, der kan t\u00e6lle og sortere forskellige typer af celler, mens immunhistokemi er en metode til at farve celler og v\u00e6v, s\u00e5 man kan se dem i et mikroskop.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><span class=\"ez-toc-section\" id=\"Hovedresultater\"><\/span>Hovedresultater<span class=\"ez-toc-section-end\"><\/span><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Effektiv internalisering og trafficking<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">T\u00e6nk p\u00e5 kr\u00e6ftcellen som et hus. Medicinen (FL1-PNU) finder den rigtige &#8216;adresse&#8217; (uPAR) p\u00e5 huset og bliver lukket indenfor. Inde i huset bliver medicinen transporteret til &#8216;skraldespanden&#8217; (lysosomerne), hvor den kan frigive sin cellegift og \u00f8del\u00e6gge kr\u00e6ftcellen indefra.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Potent cytotoksicitet<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Medicinen er rigtig god til at dr\u00e6be kr\u00e6ftceller. Den virker ikke kun p\u00e5 kr\u00e6ftcellerne selv, men ogs\u00e5 p\u00e5 de celler, der st\u00f8tter kr\u00e6ftcellen (stromale celler) og p\u00e5 celler i immunforsvaret, der kan v\u00e6re blevet &#8216;narret&#8217; af kr\u00e6ften.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Bystander drab<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Medicinen kan endda dr\u00e6be kr\u00e6ftceller, som den ikke direkte binder sig til. Det er lidt ligesom en eksplosion, der rammer alt i n\u00e6rheden, selvom bomben kun var placeret \u00e9t sted.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Tumorregression in vivo<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">In vivo betyder &#8220;i levende organismer&#8221;, alts\u00e5 i mus.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">N\u00e5r musene med kr\u00e6ft i bugspytkirtlen f\u00e5r medicinen, skrumper kr\u00e6ftsvulsten, og musene lever l\u00e6ngere.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Immunmodulering<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Medicinen hj\u00e6lper ogs\u00e5 med at &#8216;opgradere&#8217; immunforsvaret. Den g\u00f8r immunforsvaret bedre til at genkende og angribe kr\u00e6ftcellerne, samtidig med at den &#8216;slukker&#8217; for de dele af immunforsvaret, som kr\u00e6ften har manipuleret til at hj\u00e6lpe den<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><span class=\"ez-toc-section\" id=\"uPARs_rolle_som_%E2%80%9Cmaster_regulator%E2%80%9D_ved_PDAC\"><\/span>uPARs rolle som &#8220;master regulator&#8221; ved PDAC<span class=\"ez-toc-section-end\"><\/span><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">uPAR er en slags &#8216;chef&#8217; i kr\u00e6ftcellerne, der giver dem ordre til at vokse og sprede sig. Samtidig g\u00f8r medicinen det nemmere for immunforsvaret at finde og angribe kr\u00e6ftcellerne ved PDAC (kr\u00e6ft i bugspytkirtlen).<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><span class=\"ez-toc-section\" id=\"ADC-teknologiens_potentiale_i_kraeftbehandling\"><\/span>ADC-teknologiens potentiale i kr\u00e6ftbehandling<span class=\"ez-toc-section-end\"><\/span><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">ADC&#8217;er repr\u00e6senterer en lovende terapeutisk strategi i kr\u00e6ftbehandling, da de:<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>\u00d8ger l\u00e6gemiddellevering til tumorer<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Antistofkomponenten muligg\u00f8r specifik levering af cytotoksiske l\u00e6gemidler til tumorceller, hvilket minimerer bivirkninger p\u00e5 raske v\u00e6v.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Overvinder l\u00e6gemiddelresistens<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">ADC&#8217;er kan omg\u00e5 mekanismer for l\u00e6gemiddelresistens, der ofte observeres i kr\u00e6ftceller.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Inducerer immunogene celled\u00f8d<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">ADC&#8217;er kan udl\u00f8se immunogene celled\u00f8d, der aktiverer antitumorimmunresponser.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">FL1-PNU udnytter disse fordele ved ADC-teknologi til effektivt at m\u00e5lrette PDAC (kr\u00e6ft i bugspytkirtlen).<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><span class=\"ez-toc-section\" id=\"Betydning_og_perspektiver\"><\/span>Betydning og perspektiver<span class=\"ez-toc-section-end\"><\/span><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">Denne unders\u00f8gelse giver et st\u00e6rkt pr\u00e6klinisk grundlag for at udforske FL1-PNU som en potentiel klinisk kandidat til behandling af PDAC.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">FL1-PNU&#8217;s evne til at inducere tumortilbagegang og omforme TME (tumormikromilj\u00f8), \u00e5bner for nye muligheder for at forbedre behandlingsresultaterne for patienter med PDAC, enten som monoterapi eller i kombination med andre behandlinger.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><span class=\"ez-toc-section\" id=\"Konklusion\"><\/span>Konklusion<span class=\"ez-toc-section-end\"><\/span><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">Konklusionen er, at FL1-PNU er et lovende nyt antistof-l\u00e6gemiddelkonjugat, der effektivt m\u00e5lretter uPAR i bugspytkirtelkr\u00e6ft. Ud over at dr\u00e6be kr\u00e6ftceller direkte, omformer FL1-PNU ogs\u00e5 tumormilj\u00f8et til at v\u00e6re mere gunstigt for immunsystemets bek\u00e6mpelse af kr\u00e6ften. Disse pr\u00e6kliniske fund baner vejen for klinisk udvikling af FL1-PNU, enten som monoterapi eller i kombination med immunterapi, med potentiale for at forbedre behandlingen af patienter med bugspytkirtelkr\u00e6ft.
